Product name:Driselase
Product description:A crude enzyme containing mainly cellulase and pectinase, also mixed with protease and nuclease. Speed ??up the training apical cell nuclei dissociation. Collapse of the enzyme (Driselase) can be hydrolyzed cell wall (mainly fungal cell wall) of a mixture of a variety of enzymes, useful for preparation of protoplasts.
Properties:powder, Protein ≥10 % by biuret
Storage Temp:-20°C
Application:For the preparation of protoplasts1,2; Analysis of pre-mRNA processing in transfected plant protoplasts3.
Preparation method:Place 0.2 g driselase in a conical flask, dissolve in 10 mL 4 M mannitol and adjust the pH with 0.1N HCl to 5.2. Suspend 1 g of cultured cells in the solution. When the enzyme is allowed to react with the cells for 1-2 hours while gently shaking the flask in a water bath at 30°C, most of the cells used as materials lose the cell walls and become spherical protoplasts.
Unit Definition:1 U corresponds to the amount of enzyme which liberates 1 μmol glucose from carboxymethylcellulose per minute at pH 5.0 and 37°C; further enzyme activities: pectinase, xylanase, dextranase, laminarinase, amylase, protease, macerating and lytic activity; commonly used concentration: 1.0-2.5 %
